Still waiting for 100x oil objective, so this is with UPlanApo 40x on Olympus BX50, strongly cropped. The first is a Zerene stack of 6 manually shot images. I originally included both sides of the test in the stack, but the resulting stack was confusing. This is only one side of the test plus the outer wall seen on-end. DIC, with the kind of colored haloes that suggest to me the test is mineralized.

The second is much larger and less cropped, a single image showing the test surface pattern.

PS - Both subjects come from a semi-dry moss and soil sample placed in a container with water and left exposed to indirect daylight for a couple of weeks.

Arcella, from the same sample. Lightly fixed with diluted Lugol's.

Good to see some (very clean) SEM pictures of similar tests. They are good examples of how translucent objects render in completely different ways in SEM and optical microscope. And of course DIC adds its own "interpretation" of 3D structure.

I wonder how they would look dry in incident illumination at 100x or so (without DIC) with a Mitutoyo M Plan Apo 100x or similar objective. Unfortunately at present I can only get up to 20x with these objectives.
