FOV focus stacking

Hi there,
I now got my Mitu M Plan APO 5x and done some shots with it.
Absolutely happy with my insect stacks. My mineral stacks need improvement.
On the Mitu spec it says depth of focus is 14µm. The focus stacking calculator on Extreme-Macro says 31µm as I convert the NA 0.14 to f/# as 3,53. I think if I get the StacShot and set it to 10µm steps it would be fine?
Currently I now move manually 25µm....

Yes, this is 5x with Canon EF 70-200 f/2.8 set to 200 on my EOS 7D

Mitutoyo is quoting single-sided depth of focus, that is, maximum deviation from perfect focus.

Total depth of focus is twice that, so 28µm using Mitutoyo's assumptions. This is only a little different from Extreme-Macro's calculator, due to slightly different assumptions.

Your current step size of 25µm is fine.

Reducing that to 10µm may make things even better, assuming that more than doubling the number of frames is not a problem for you.

Thanks for your reply.
The numbers of images is no big deal really. I´ve tested 600, 16 bit full resolution tif images from my 7D. Works like a charm.

I´m considering StackShot to improve my images....
