Hi Litonotus,
Thank you,

I just put a few drops of alcohol 70° into the water directly on the slide with the daphnia. If there is enough water, the daphnias stay as they swim.
Few minuts after, you see the heartbeat decrease slowly. I think they fall into a kind of ethylic coma.
Do not put any slide cover, but when I take photos, I just avoid to make any vibration or to breath too heavy

I have sometimes just breathing while working in a thicker aqueous environment can jostle the specimen with or without a cover glass. In those cases I tape a piece of paper to the turret that extends just below the stage.
I also keep pure grain alcohol in a bottle at hand to make micro crustacea "happy drunks."

Very nice pics! Does Magnesium chloride work as well to "anaesthetize" them? I used to routinely use mag chloride for working with echinoderm larve

Pure grain alcohol, whisky, gin or vodka ?

Francis, I already used Magnesium Chloride, but I think alcohol is faster.
Perhaps I have to try one more time with another concentration

Nice picture Jean Claude,

I saw before similar pictures from Julio Moreno but in darkfield
(look at https://www.flickr.com/photos/rogeliomorenog/ )
I am happy to find the secret for fresh but unmoving Daphnia: to kill them with alcool!
but after you need not move the water for 65 pictures!!

There are somme missing details in this picture ( specialy on 2e): some bristles on edge of shell are not apparent, or the surface of the shell is not complete.

I don't know if it is because you don't take enough pictures or because focus stacking software can't work well with external and internal diascopic details?

Perhaps, can you show this picture on french forum.
That will be easier for me to write in french!

But in this topic, could an expert give us an answer, and tips to focus stacking in transmitted light with complicated objects?

Hi,

I have checked all the pics, and it seems, that it is the software that sometimes didn't know the good clean parts to choose. In fact, all the détails are present, but the software doesn't understand between the inside and outside part which one to keep.
