Did you happen to know what the internal granules are which are highlighted by the POL image? Possibly starch?

Thank you, the magnification on sensor was 40x (20x objective x 2x phot lens on the front port = 40x)

Can you please give more details of the "2x phot lens on the front port"? I wonder how this approach would compare to using a 40x objective and direct projection?

Des, it is a 2x photo lens that came with my TE300, looks like it is a Nikon standard to put 2x or 2.5x photo lens at the front port of the inverted scopes, the front port is normally used to mount SLR camera. Iw ould like to have a digital full frame to mount it on the front port of my TE300. The Plan Apo 20x has a numerical aperture of 0.75 (I normally use it at lower NA closing the condenser diaphr. to balance, to my like, resolution and contrast).
