Thank you very much all for your comments.

The last image in full resolution and tif quality is like Litonotus said: without words.

To get bright and shiny image try the following:
- Avoid to shoot with too much water between the subject and the coverslip.
- Try to select the coverslip that you use. I use #1 coverslip and I try to select the ones with 155um or more (I use a Mitutoyo micrometer with 1um resolution), normally with a coverslip of 160um (you have 10um of play) you get very nice image quality.

Let me know if you need more details.

Beautiful image with very rich surface detail. Are you using a contrast or detail enhancing step, other than simple sharpening, during image post processing?

The image on the camara's display look beautiful; but when I convert the raw to tif it is not as on the camera's display so I have to adjust contrast. Normally I do the following:
1. Convert raw to tif.
2. Rotate and crop.
3. Remove any unwanted debris on the background.
4. Adjust contrast.
5. Sharpenning.
6. Remove noise with DeNoise.
7. Save tif.
8. Resize and save jpg.
