Impressive!

What is the specimen, and how was it prepared?

Hi Rik,
Many thanks!
It is Psylla buxi in euparal.

It looks to me like muscles in the legs are still intact. Is that an illusion, or are they really?

What was the preparation before the euparal?

Hello Rik,
Can it be that it is due to the lighting?
The oblique illumination was combined with reflected light.

Likely cross pol would clarify it

Nice image, BTW!

To clarify my question...

When I asked, "What was the preparation before the euparal?", I was hoping to learn what sequence of chemical baths the subject went through.

Hi Pau,
OK, I have just checked it.
The image looks similar, except that the muscles are snow white.
Is that what you meant?

Hi Rik,

Not much chemistry was used :-)

They were first swept from the boxwood,
briefly fixed in AFE and then bathed in Isoprop 100%.

Then on the slide, let the alcohol evaporate a little, Euparal and cover glass on it.

AFE = 5% Acetic acid + 5% Formaldehyde + 90% Ethanol

Yes, I suppose. Because muscle cells are birefringent due to the parallel protein fibres they are highlighted with cross pol, often with strong interference colours.

Hi Pau,
The muscles appear rather white in polarized light:

The muscles appear rather white in polarized light
and very well rendered, I can see the periodic bands striations ,at different places like at the upper right corner.

Interference colours are not typical of a given material but dependable of the thickness together with the RI and the direction of the light. According with the Mitchell-Levy colour interference chart, low 1st order colours are pale grey, so they will appear white against a black background.

Hi Pau,
Thank you for the hint about the interference colour chart.
