It looks to me like muscles in the legs are still intact. Is that an illusion, or are they really?

The image looks similar, except that the muscles are snow white.

Hi Rik,
Not much chemistry was used :-)
They were first swept from the boxwood,
briefly fixed in AFE and then bathed in Isoprop 100%.
Then on the slide, let the alcohol evaporate a little, Euparal and cover glass on it.
AFE = 5% Acetic acid + 5% Formaldehyde + 90% Ethanol

Yes, I suppose. Because muscle cells are birefringent due to the parallel protein fibres they are highlighted with cross pol, often with strong interference colours.

and very well rendered, I can see the periodic bands striations ,at different places like at the upper right corner.
Interference colours are not typical of a given material but dependable of the thickness together with the RI and the direction of the light. According with the Mitchell-Levy colour interference chart, low 1st order colours are pale grey, so they will appear white against a black background.
