Hi,

In the last few weeks I could not expend much time with scope and when I did I did not manage to get results worth sharing.
A new stack, adapting to the microscope is proving to be quite challenging. Preparing the samples and working with live moving subjects.

It took me several times until I could complete a stack of this Diatom (ID?) from a marine water sample, it reacted a little to polarized light so I run this one. 22 shots with nikon CFN Planapo 40/0.95 160/0.17; direct projection plus 50% crop with Sony A5100

Until now with every diatom (or almost anything else) I have tried to run a stack with I had to retouch from individual frames, no way I can get a clean stack straight on. Deciding what details to take from each frame is difficult as I do not fully understand yet what I am really looking at and which details are important to keep.

A different polarized stack, this time I left the backgound blue, which seems to suit it better than trying to fix the WB

and one trying to keep only superficial texture, more translucent like. There seems to be some banding, will try to fix it

Well, painful as the workflow might be the results are hard to argue with. Great work!

Seems like a pleurosigma indeed, forgot to mention it is from a marine water sample

It's a resolution chart! The small pores pose a major challenge, the are hard to resolve.
