Lilly pollen, Nikon BD Plan Apo 150x NA 0.90. For this picture I used darkfield light. The high NA makes every dot, spot, line... on the sensor visible so I did some retouching both before and after stacking. I darkened the background and sharpened the picture. Canon 5D II, Zerene Stacker, Adobe LR and PS.

So, this was illuminated with light coming up through the specimen?

It is a Bright field/Darkfield (BD) EPI-light lens so light from above.

Really beautiful! These kinds of pictures would be great tools for studying pollen samples when the species is unknown, especially ancient pollen.

Hi Saul, I used a Nikon Optiphot microscope. The microscope have a 2.5x projection lens, so a lot of empty magnification and 375x on sensor. The 150x lens only have 0.3 mm working distance.

In this case I wanted to show the colours of the pollen grain. For ”clean” black and white pictures I think that a GIF (green) filter is a good option.
