Bon Jour Olivier,

Excellent photos, but the staining is weak. I have had this problem with ciliates. Sometimes stains work well and other times - sometimes with the same species - hardly at all. Lugols Iodine and methyl green have always been the most reliable simple stains for me, but still I have not been happy with the results. Sometimes I have had poor staining because of the medium that the cilate is in - for example - sea water.

It would be nice to hear what everyone out there thinks.

Well, I have neither heard of Lugol being used for nuclear staining nor of using it simultaneously with methyl green!

Usually methyl green is used acidified with acetic acid or in combination with pyronin. Also orcein acetic acid is being used. The color sort of develops or rather changes throughout the process, it is best between 2 and 5 min. Before that it will be to faint and after a while it will turn darkish.
