Hypotrich ciliate

Post by Charles Krebs »

About 5 weeks ago I let a sample dish go dry. It sat, bone dry, for about a month. Out of curiosity I added some water (and also some water in which I had boiled some cabbage leaves). With about 24-36 hours it was teeming with bacteria and there were quite a few of (apparently) the same, very active, hypotrich ciliate seen earlier.

Olympus BHS, 40/0.95 S Plan Apo, DIC, Canon T3i, electronic flash.

Amazing details! Thank you, Charles!

This ciliate seems to be a naturally low contrast subject.

Did you pick the dark grey color to enhance contrast?
Did you have to close down condenser or objective iris?
Did you immerse condenser?

Yes a clear subject, no color at all. This was with DIC, so the "slider" was adjusted to provide the best contrast, which made the background darker. I converted to b+w, as I occasionally do with completely colorless subjects. The condenser was closed down just a little, but not oiled (my BH2-UCD DIC condenser is a 0.9 NA and can't be oiled).

Very nice. I would be interested to see a color and B&W version for comparison, though the B&W is very effective.
