Daphnia Details in Darkfield

Post by Charles Krebs »

Sorry... just had to use the alliteration in the title

Unfortunately not much time on the microscope lately, but I did come across some daphnia. I wanted to do these with darkfield primarily because of the eye. With brightfield or DIC the middle part of the eye becomes primarily a dark, detail-less "blob". With darkfield you can see much more interesting detail. Live specimens with electronic flash, 10/0.40 and 20/0.70 objectives. Olympus stand with Nikon 0.95-0.80 dry darkfield condenser.

They are beauties and I hope you will be able to find more time for microscopy!

They are all great shots, but my favorite is the final image, with the delicate glass-like branches. I wonder if they are birefringent, i.e. how would they look with POL?
