Collection of Pediastrum from the pond right in front of Janelia institute. I used the same technique to stain the cell wall (Calcofluor White) and image the specimens (405, 488 and 562 nm laser lines); the variations in the appearance of individual colonies, or even individual cells within them, was a bit of a surprise for me.

Imaged with Zeiss LSM 710, 40x NA 1.3

Images are spectacular as usual. These are unfixed right? I wonder if the variation in the fluorescence is due to some of the segments being alive or dead or in between. I've seen that in some Pediastrum. I must admit I am jealous of the Janelia pond. My pond has a lot of Cymbella and other diatoms in it, but not so many desmids in the winter.
