hello Michael,

great to see more plant anatomy slides from you - and looking forward to seeing your own sections.

These photos are captioned the wrong way around:

top (pink) photo is the root.

and bottom (blue) is the stem.

thanks for posting,

Brian

Zea sections

Post by Olympusman »

Dang, they are criss-crossed. Thanks, guys.
Aquestion about mounting sections -- I found a recipe in the Microtomists Vade -mecum for a mounting medium using egg white and Sodium salicylate to "glue" the section to the slide for dissolving the parafin with glycol and then doing the stains. Does this need to be refrigerated or does the salicylate render it sterile? Thanks ahead of time.

hello Michael,

this comprehensive article more or less answers your question (towards the end of the article):

http://www.microscopy-uk.org.uk/mag/art ... para2.html

He uses thymol instead of salicylate... but I doubt there is any difference.

I think this is the best article on making sections for amateurs that i know.

good luck... I am looking forward to seeing the results,

Brian

Microtome sectioning article

BJ,
I have been familiar with this article for some time. What is frustrating is that while the first two articles are very thorough, he never finshed the series with the third article about clearing the parafin and staining.
