Hi David
I use formaldehyde to fix them: 3 drops in 5 ml of ciliate sample
Rinse Twice in distilled water (using centrifugation)
I then add Pyridine and peptone solution
Then I add silver carbonate solution
complete with 20 ml water
heat at 60°C
Chill stop the reaction (turning brown) or use a silver fixator (hyposulfite)
Rinse and centrifuge twice
pick cells individually and observe under the microscope.

your work is excellent! I am trying to impregnate with this method (modified by Augustin, Foissner and original too) astome ciliates but its very weak and bad. Please can you tell me your exact recipe to Rio Horteg solution or can you give me some advice? I´m confused because its the most important taxonomic character :/ Thanks a lot!!!!
