Closterium: cell division in time lapse

Post by myriophyllum »

Hi,

I'd like to show one more time lapse video now.
It is the third part of my little series of desmid divisions -
after Micrasterias and Euastrum here a Closterium:

http://www.youtube.com/watch?v=VtuBmXl0ttM

The cellular surrounding has in one scene been pointed out by putting some chinese ink into the water.

Illumination: Darkfield and asymmetric Rheinberg/Col mix
Zeiss Standard 18; Neofluar 16; Planapos 10, 25, 40 and 63; Ultrakondensor; Achr.-Apl. 1.4.
Time lapse (up to 250x) using the MagicLantern extention for Canon EOS.

My other desmid videos can be found here:

Best wishes

Jens

Hi Tom,

no, RAW video doesn't work for Full HD on my 5DII, unfortunatally.

Thanks for your compliment

I love videos that show a story and a theme. So your videos of the desmids are fascinating and satisfying for me to view. Your work is not only artistic, but it is also show great technical mastery over your equipment.

How do you make your slide mounts so that the plants can be maintained for these prolonged video sequences. Also, do you have any problems with focus drift when taking time lapse?

Konstantan spacers

Post by myriophyllum »

Thank You Brian,

I don't have much problems with focus shifts (more with the movement of the desmids...).
For constant distances between coverslip and slide I'm using Konstantan wire as spacer. This wire is available down to 0.04 mm diameter, here I used the 0.150 mm. From time to time it is necessary to replace evaporated water...
It helps to use large 24x40mm coverslips, too.

Best wishes

Jens
