Pollen grain

Post by Linden.g »

I came across these two pollen grains on the wing of a Protographium agesilaus butterfly. Imaged with ULWD Neo S Plan 50x and NFK 2.5x. Lighting from the sides no direct epi lighting.

Very nice. I like the contrast between the spherical pollen and the flattened scales of the wing.

Very interesting and beautiful images !

What is the working distance of this objective ? I am going to buy some objectives for epi micrographs. Could you give me any advice if about such objectives ? I think about 20 X and 40 X ones. Which of such objectives are the best for epi photomicrographs. I am going to use Jansjo IKEA lamps.

Thanks for the kind comments.

Marek, I've only just started to use this type of objective. The working distance is 8.1mm which is great to work with for side lighting but with the downside of reduced resolution due to the lower NA of 0.55 compared with the standard 50x neo S plan at NA of 0.8. These are infinity corrected objectives which need the correct tube lens (or 200mm telephoto), I'm using the BH2-UMA epi unit. If you go with true epi bright field you will need the cross polarization filters due to back scatter and low contrast. You don't need polarization with side lighting. If you are only using side lighting and not epi using the UMA on a microscope there are probably better long working distance objectives which can be combined with a telephoto lens which other members of the forum are more familiar with.

If I may add my 2c, I purchased the Olympus 20/0.45 and 40/0.6 LUPlan objectives. The correction collar is most probably the single most useful aspect of these objectives. Their image quality is also incredible.

Waldo,

Thank you for your info and advice.
By the way, could you attach the photographs of your mentioned objectives ? I was trying to find them in Google but I did'nt managed it.
