Two contrast enhancing techniques

Post by Brian Matsumoto »

I thought the group would be interested in the altered appearance of diatoms when their contrast is enhanced by two phase techniques.

One of the images is from a Reichert Anoptral contrast objective that provides a sepia toned background. It was advertised as having the advantage of not generating phase halos. This does not seem to be the case in the photograph. The other image was taken with a Heine Pv objective. One can see that the background is more of a cold blue tone.

The two objectives provide a differing view of the same subject and, aesthetically, I am preferring the Anoptral image. The different magnification is due, in part, the Reichert is a 45X objective and the Heine is a 40X.

What do you think?
