Jacek, again excellent pictures.
I'm very interested in your fluorescence equipment, as I'm setting up a similar one with the same epi illuminator/condenser. Please take few minutes to answer me some technical questions
- The Zeiss Standard fluorescence condenser is coupled to another kind of microscope (a PZO?). Does it couple natively o did you modify it?
- Do you use a white LED or a blue one?
- What expossure times are you getting with this LED?
- Could you explain the fluorescence cube writings?:
450-490
FT 510
LP 520
I assume that they are referring to wavelenghts, but they are pretty close...

Thank you for your nice comments
Pau, FL only knows, I learn.
Yes, a set of Zeiss FL is connected to the microscope PZO Biolar.
Fit together almost perfectly.
My 10W LED is not suitable as a light target, I have to use something else. The LED has nine tiny points of light, it uses only one.
White LED is a cold light
ISO: 80 - time: 1-3 s
In general, lack of light, I have to look for organisms with a strong autofluorescence.
I'll try to use another LED and halogen 100-150 W
It is very important aperture lenses. The higher the better.

Jacek,
These are really nice. I see that you have perfected the use of your UFG for high magnification objectives now. Has this involved a redesign of the filter?
You really have managed to find a low cost alternative to DIC which, on your 100x objective examples, looks just as good as your DIC pictures.

Nice images!
Pau,
these cubes contain 3 elements: the excitation filter, the dichroic splitter and the emission filter. 450-490 means the the excitation filter will allow light with wavelength 450-490 nm to pass. FT stands for Farbteiler (color splitter), that is the dichroic splitter. It will prevent light with wavelength under 510 nm to pass. LP means long pass and light above 520 nm will pass.
Regards
Ecki

Peter, I UFG original filter, made by the author - a friend Litonotus.
Filter does well on strong lenses (40, 60 and 100 X), but only if the product is well made and as thin as possible.
Ecki he needs cube to get the blue fluorescence ?
What to look for, what signs? ?
second photo Botryococcus /ob. 100 X/
