Here two specimens that I found in my sample from lake Gamboa.
20x objective (I should have used the 40x with the M. truncata but then the step between frame has to be 0.5 um, maybe next time).
Auto-fluorescence with ultraviolet illumination.

Yep first one is brilliant. Wonder what you will do once your trial expires? Mine has both ran out of time so I am back to normal processing.

I was meaning to ask - the cube you use - what are the wavelengths? Same as those I gave you? Because mine cuts off blue light so if I want that I have to composite DAPI and the custom cube.

Waldo, my cellSens and Autoquant ran out, now I have Hyugens for 30 days.
I am using the Nikon UV-2B cube, you can find the info on the following link:

Ultraviolet Excitation Filter Block UV-2B Specifications:

Excitation Filter Wavelengths: 330-380 nanometers (bandpass, 355 CWL)

Dichromatic Mirror Cut-on Wavelength: 400 nanometers (longpass, LP)

Barrier Filter Wavelengths: 435 nanometer cut-on (longpass, LP)


I hope to have soon the funds to order the filters with the wavelenght that you gave me.
