I finally got a stepper motor (same as the one on the StackShot rail) in the mail, connected it to an old Optiphot microscope and took some pictures. The microscope is much less sensitive to vibrations than my copy-stand – but there is not much room for light and posing the subject.

For these pictures I used two led lamps and diffuser. The first picture is a Sporophyte capsule full of spores, (Nikon plan apo 4x 0.2). The second picture is a section of the underside of a leaf from a Stinging nettle Urtica dioica (Nikon 20x 0.4 LWD).

Hello

Interesting photos, the sharpeness is good.
lighting is not strong enough.
May i suggest you to try your stack with one flash, diffused by one or two yoghurt pots.

Thanks for your comment. I looked at your Peristorm pictures – truly inspiring; www.photomacrography.net/forum/viewtopic.php?t=19330

Flash was a good idea and turned out to be necessary for larger magnifications - the sporophyte moved a lot by itself.

Here are two pictures, flash, Zerene Stacker, Slabber Jockey, Nikon 4x plan apo 0.2 and Nikon CF plan 10x 0.3.

Lighting is better.
Sharpen is also amazing.

Another suggestion : clear blurring around some details

Fredlab, thanks for the demonstration, I realise that I have a lot to learn about post-processing.

Here is a final picture, cross-poralized light. Some retouching to get rid of spots and hot pixels. Little curve and sharpness adjustment done in Canon DPP.
