Ixodes - Polarized light + stacking + panorama

Post by Fredlab »

Hello

I have not had much time this summer to post some of my works. I spent my time doing panoramas on thin sections of rocks (I will show it in other threads).
I took the opportunity my microscope is in "polarized/analized light mode" to see what gave few biologic slides.
A tick, prepared by a friend, gave a spectacular result.

Image

Striated muscles appear clearly.
In larger size - here

About technique used : 12 files of 50 images stacked (SPlan 10 + NFK 2,5 + EOS 550D), stitched with AutoPano.
The original size of this panorama is 16000 pixels length.

Excellent pictures. I especially like the first one. Your friend has done a good job of the preparation and mounting.

I am very impressed with this series. Ticks are often very dark and with quite a hard carapace, making them difficult to mount. The usual methods to clear and soften them involves alkali which dissolves muscles, leaving a less interesting specimen.
Perhaps your friend was lucky to find a tick which did not need this harsh treatment, but I would be interested to know his method of preparing this specimen if he was prepared to share it.
