Blood

Post by Fredlab »

Hello

Yesterday, i tried my DIC on pig blood

Image
(Leukocyte - granulocyte)

My equipment has been pushed beyond its capabilities: BHS 100W powerfull - SPlan 100 - Canon 800 isos...
These picts are noisy and not very sharp...
I'm thinking about flash to solve the problem.
I apologise for my poor english

To be honest I think those pictures are pretty good and you will struggle to extract very much more detail from blood leukocytes (white blood cells) which are really very small. Surface blebbing, internal granules and nuclei are all very clear in your images. Sure a X63 or X100 high numerical aperture Planapo might do slightly better, but the difference would not be huge. Flash may help to cut the ISO 800 down but it won't give more detail by itself. At ISO 800 your images are going to show some noise and noise reduction is essential. The trick is to remove most of the noise without destroying too much detail. It's a trade off, but good noise reduction software like Topaz DeNoise 5 or Noise Ninja will give you plenty of sliders to optimise the balance. After noise reduction, a very small resharpening may be needed, much as you might do after reducing an image to Web size.

There is little true colour in an image like this so taking out the yellowish green cast and the little bit of blue/magenta fringing will also help with the final appearance of the image.

I have made these small changes to your original for my own interest and I must say if I had taken the final image I would have been pretty pleased. :)

Hello

Many thanks for your complete com'.
If you want, HERE, you can find the original size... after my works with photoshop (changing WB - sat/contrast + denoising with Neat Image - resize, sharpen...)
One of "raw" shot is HERE.

All members can play with these picts and they can post their result in the thread.

Looking at your 'raw' image it looks like you did a pretty good job already! Anyway out of interest and with thanks for allowing it, here is my edit of your original large composite. Basically I did what I said in my other post. Noise reduction with Topaz DeNoise 5 then a bit of smart sharpening with Topaz Detail 3, desaturation of the yellow, green and blue areas with Photoshop and resize for web. Hope you like it. I have tried to retain as much detail as possible while cutting noise.

Hello

I agree - your picture is better than mine.
Thanks.

(i know DeNoize and Noise Ninja, but for my Photoshop of my Mac, i bought Neat Image, less expensive (?))

It should be possible to get very good results from NeatImage. It is a question of experimenting with the settings which preserve detail against those that remove (blur) noise until you get exactly the result that satisfies you.
