Amphipleura pellucida, part 2: the highest resolution.

Post by myriophyllum »

Hi,

in the first part of my Amphipleura posts (http://www.photomacrography.net/forum/v ... hp?t=19501) I presented some images using darkfield.
Let me now show the best resolution I could reach.
Again I took my Leuchtbildkondensor (nA 1.5) and stopped down an objektiv 63/1.4
to about 1.2 asymmetrically by a black paper.
Two crossed polars improved contrast.

First an image using white light:

blue light, 450 nm; filter 450DF50:

blue light again, but focussed slightly deeper:

I think the different look of the outer and inner side of the shell produces that effect.
For a comparation: on the sites of Peter H. You can find this very fine image:

finally, I have some distances:

Zeiss Standard 18, Planapo 63/1.4; S-Kpl 16x/12.5; 63mm relay lens; DSLR
I collected the diatoms at the Kraterquelle of Bad Nenndorf, Germany.
Naphrax was used for embedding.

Your comments are welcome!

Greetings

Hi,

just for comparation:
Amphipleura pellucida under a REM,
Peter took samples from the same pond to make these images.

That's some resolving power! =D>
Near the calculated limit for 1.4 NA objective (0.24 μm with green light) - that is for l1 line in your photo, l2 is discussable if resolved
