Amphipleura pellucida, part 1: darkfield at the limit.

Post by myriophyllum »

Hi,

I've tried to resolve as many details as possible of the diatom Amphipleura pellucida using darkfield.

So a very high nA (Carl Zeiss Leuchtbildkondensor nA 1.5) oil immersion DF condenser was combined with a objective 63/1.4, stoppt down to about 1.3
by a black paper with a hole in it.

First an image using white light:

blue light, 450 nm; filter 450DF50:

Zeiss Standard 18, Planapo 63/1.4; S-Kpl 16x/12.5; DSLR

I collected the diatoms at the Kraterquelle of Bad Nenndorf, Germany.
Naphrax was used to embed the shells.

Your comments are welcome!

Greetings

Jens

Jens, nice exercise breaking the limits!
Were do you place your paper stop in the Planapo objective?

Thank You, Pau,

simply onto the upper hole, on the plastic part, that is screwed in there.
I know, a better position would be inside the objective... ;-)
