Very nicely done, much better than I managed at college and very well photographed. Your pupil deserves an A* for the root tip squash. :D

You are right.
My pupils are aged 16 (lycee...)

Protocole is easy

Young root end - put on a slide - 5 to 10 min in one drop of HCl - Soak acid - one drop of acetic orcein - 15 to 20 min - squash.

What is that structure in photo 4, the horizontal lines in the upper right of the photo? I found similar structures in the skin of an onion? Pardon my ignorance, I never took biology past grade 9.

Hello

I'm not sure @ 100 %, but i think it's some cristals of Calcium Oxalate.
With google, we can find "raphide" for these structures.

Superb - wonderful way for your students to learn their biology lessons, especially with the immediate, tangible visual result. They aren't likely to forget such a rich experience.

Please note, this is a 9-year-old thread, resurrected by new poster Sm55 to ask for help on a technical issue.

Sm55, your question would be better posted in a different part of the forum, either "Macro and Micro Technique and Technical Discussions", or perhaps better in "Beginners Micro". When you do that, please be more specific about what you need. Do you need a general tutorial on DIC (Differential Interference Contrast), or guidance about a specific piece of equipment, or something else?

I want to do DIC imaging in a brighfield setup. I want to test a set of permanent mount slides.

In that case, I suggest to start by asking any internet search engine about something like differential interference contrast tutorial.

At this moment Google returns to me several YouTube videos that appear relevant, plus https://www.scientifica.uk.com/learning-zone/differential-interference-contrast and other pages of non-video material.

Note that DIC requires specialized hardware in the microscope. If you have access to such a microscope, then please tell us what make and model it is, because that may affect whatever advice you get.
