First heliozoan under my hybrid DIC microscope. Leitz NPL Fluotar 50/1.00

1-Single shot

2- Stack attempt, many artifacts as the beast was slowly moving and the pseudopoda bobbles don't stop traveling (and both Combine ZP and my poor stacking skils may not be the best combination). The amount of bobbles showed is much higher than the actual one due of its movement. but I post it because it may give some idea of the amount of spiky pseudopoda

Anything that moves is tough to stack, but your result here looks great to me! I think the two pictures together give me an excellent impression of how the beast is built.

Hello Pau,

you are right: sometimes it is good to make experiments, because the DIC lenses are terribly expensive. But excuse my curiosity - I dont know what is a hybrid DIC?
An other question: is the Leitz NPL Fluotar 50/1.00 thought for Oilimmersion and for a tube lenght of 170 mm? Did you use oilimmersion?

-I call hybrid my microscope because I mix components from different makers:

-Yes, it is an oil inmersion objective and I use it oiled, but corrected for 160 mm (altough for this magnification, according Leitz literature, a 170mm corrected one would be equally adequate)

Pau,

You are getting a good DIC effect.

These are always frustrating to photograph because when viewing, you can clearly see the large number of axopodia but the limited DOF really makes it hard to show this nicely. And if the specimen is flattened you lose the wonderful 3-dimensional characteristic.

(It might be a fun experiment to "focus through" while taking a video and pull out the frames and do a 3D rendition using Zerene stacker).

The video stacking trick would be a very good idea for slowly moving critters like this one.
