Euplotes eurystomus and Pinnularia (images added)

Here is a set of Euplotes eurystomus images and two diatom images.

All of them with Nikon TE300 microscope, Plan Apo 20x/0.75 for images 1, 2, 3, 4, and 5; Plan Apo 40x/0.95 for last image; DIC, flash and Canon T3i mounted on the front port of the TE300.

Dimensions:

- Euplotes:
pic. 1: 157 x 69 microns
pic. 2: 165 x 64 microns
pic. 4: 133 x 65 microns

- Diatom 140 x 21 microns

congratulations on finding Nikon CFI plan apo 40, it is extremely rare I have never seen it on ebay...

I was lucky because at the moment I asked an ebay seller he was about to list it on ebay.

Top pictures as usual.

The last Euplotes seems an snake or even a pelicosaur head ready to bit. The optical sectioning with this high NA is impressive.

Hello Rogelio,

I am very much impressed by the quality of your diatom pictures! In order to better understand your method (and maybe to improve my pictures) I would need some exif-data, for exemple: shutter speed, exposer mode (manual?), live view, what kind of flash etc.

Absolutely excellent pictures. The diatom is outstanding. Another top class lens, I hope you didn't have to take out a mortgage to pay for it . :lol:

I agree with you, I am very happy with the TE300 + High NA kit + 0.85 condenser lens + Plan Apo objectives.

The following is my normal configuration:
Shutter speed: 1/200
Expouser mode: Tv
Mirror lock up, no live view.
Nikon SB-80 flash in manual mode: with the 20x objective I normally shoot at 1/2 power or less (when the camera shows that I am clipping the highlight). With the 40x at full power. I have to look for a more powerful flash if I want to shoot with 60x (I do not have it) or 100x and the camera mounted on the front port of the TE300 (the front port has a 2x photo lens).

Actually I am using DIC system that is used on Nikon 80i/90i/Ti; but I did a test with DIC system that is used on E600/E800/E1000 and no differences in image quality were found.

One point to mention is that my Senarmont polarizer and analyzer are both high transmission, from the following link:

I quote the following paragraph:

"The polarizers employed in DIC microscopy are similar or identical to those used for polarized light observations, but many manufacturers offer matched DIC polarizing filters that have both low and high light transmission efficiencies. At the upper end of the magnification range (40x, 60x, and 100x), polarizers with high transmission efficiencies are preferred for DIC because specimen details are more clearly delineated and larger bias retardation values can be utilized for video and digital imaging. The low transmission polarizing filters are useful for DIC observation using the 10x and 20x objectives, but severely restrict light transmission at higher magnifications."

I could deal a very good price for a Plan Apo 40x almost new (used as demo 2 or 3 times).

Here two images showing a close-up of the Pinnularia´s picture taken with the Plan Apo 40x/0.95.

Look the details captured by the 40x/0.95, the separation (where you can see to the other side) that you see on the top row are around 0.4 microns!

I do not have a Plan Fluor 40/1.3 oil to do the test. :(

Franz,

I forgot to mention:

- White balance: Auto
- The SB-80 has a zoom that normally I set to maximum (105mm) to get the maximum light from it. When you are shooting at maximum power the next lower power is 1/2, instead of doing that big change I lower the zoom step by step until the clipping of the highlight disappears.

Hi Rogelio,

magic images! Is this correct: the Nikon TE300 is a inverse microskope? I never saw DIC-pictures with a inverse microskope like this.
