Last time I struggled with my Nikon E200 100x objective in oil. Same setup, just a fresh attempt. I had a brine shrimp that died in some water, and these bacteria started growing exponentially. Taken under the 100x oil objective under Phase Contrast.
This looks very good for phase contrast with a 100X. Bacteria of this type are a tough subject with a "light" microscope.
I think you could boost the contrast with a "levels" or "curves" adjustment, and add a little unsharp mask and it would look a bit better and not over-processed.
Nice pictures. I also made some bacteria photo's. I let them grow on a petri-dish with Agar, ented with some pondwater.
In the begin there was a lot of moving but after a few minuts they where all static. A Plan40/0.65 and 2.5 photo-oculair used. Kreutz filter for oblique light.
