I tried around five times to take pictures, when it did pause, to do a stack of this specimen.
Nikon Plan Fluor 40x/0.75, DIC, Canon 50D, ZS DMap.

You can probably get DMap to give an even better output if you push the contrast threshold farther to the right, so that featureless background goes all black in the preview. As it is now, you have some typical blotchy halos near the specimen that are the result of DMap trying to fit weak "detail" that isn't really detail at all but rather just OOF edges sweeping across the field.
