Here a stack of 13 frames with ZS (Pmax, no retouching).

Nikon E400, Plan Fluor 40x/0.75, DIC, flash, Canon 50D.

To get that colors I used a lambda plate (first order red) on the DIC polarizer.

Very fine image, nice clean stack. I never tire of Diatoms. They provide so much scope for trying new techniques and perfecting photography and stacking methods.

Yesterday I received my Nikon plan fluor 100x/1.3 oil objective and I could not resist the idea of testing it.

Here a stack of 18 frames with ZS (DMax, with retouching from PMax).

Nikon E400, Plan Fluor 100x/1.3 oil, DIC, halogen illumination, Canon 50D.

The improvement in resolution is striking. I guess this should be no surprise, going from NA 0.75 to 1.3, but still very gratifying to see.

No trouble with boredom at this end!

Excellent!
If you invert the image as I did here:

the frustula holes will show as holes.

I do not know if it is an optical illusion; but looking at the bottom edge it looks like the frustulas (columns) are some kind of scale (I see like one is over the other and on), any expert on diatoms that can confirm this?
