After about 6 months that I decided to sell all my finite Nikon microscope items to upgrade to infinity DIC, today I could take a partial DIC image with a Nikon CFI60 Plan Fluor 40x/0.75 (I say partial because the objective prism that I used for the test was the PA/PF 100 oil instead of the PF 40).
The equipment was:
- Custom modified (to accepts DIC nosepiece) Nikon E400 frame.
- Custom build Senarmont polarizer.
- Custom modified Nikon Turret Condenser for Ti (Inverted microscope), I had to modified this condenser to use it on the E400.
- Nikon DIC condenser prism 0.5 - 1.0 NA for oil lens.
- Nikon CFI60 Plan Fluor 40x/0.75
- Nikon DIC objective prism PA/PF 100 oil.
- Nikon rotatable analyzer for E400 / E600.
- Nikon infinity trinocular head - direct projection.
- Optem Optizoom at 2x - mounted between the analyzer and the trinocular head.
- Canon 50D camera.
The final image on sensor was about 80x (40 objective x 2 Optizoom).
The test slide was Lyrella lyra, single shot:

To be honest the picture was taken yesterday morning, before leaving to work, it took no more than 5 minutes, the test slide is a prepared slide by K. Kemp. I stayed up all night reading some chapters of the book "Fundamentals of Light Microscopy and Electronic Imaging" by Douglas Murphy, I like to read and learn how things work.

Excellent work. That looks really high quality DIC. Did you convert to monochrome because of any chromatic aberration issues or simply because the subject is enhanced by it? I would not have been confident that these components would work together at all. DIC has a reputation of being very intolerant of 'mix and match' setups.

That must have been an anxious moment. I'm so glad it worked for you in the end. It would be nice to see a photo of your set up with the various components labelled at some point, as it's hard to picture, and it sounds like a really innovative piece of work.
