Fern (Nephrolepis) stem section

Post by NileRed »

Hello,

the following pictures are an autofluorescence (blue light excitation, green barrier filter) of fern-Nephrolepis (the popular room fern) handmade stem thin section.

First picture data: Panorama of 4 pictures (Photoshop CS4), Obj Carl Zeiss Jena 6,3x Apo, iso 200, exposition time 15-20 sec, all 4 starting pictures were stacks of 10 pictures (Zerene).

Second picture was just L and b inverted channels of the first one.

Third picture data: Obj Carl Zeiss Jena 16x Apo, iso 200, exposition time 15-20 sec, stack of two pictures (Zerene).

The pictures were obtained using simple equipment shown in my previous post (Geotrichum Candidium), but without any condenser.

Beautiful images. So this is with no staining? Not sure what autofluorescence means.

Fascinating stem section, beautifully presented!

I also enjoyed reading the thread mentioned in your post, Geotrichum candidum:

Thank you for the comments.

You are right, I have used no dyes, but definitely brightness was increased.

With best wishes
