Euglypha brachiata

Post by Charles Krebs »

I collected some mosses and mud from a small stream where I have always found testate amoebae present. This is a species I really like, and I must have spent about 3 hours just looking for one and getting it onto a slide.

Taken with Olympus BHS, Olympus S Plan Apo 60/1.40, DIC illumination. 41 image stack. It is very transparent, so stacking was done in 4 sections to retain surface detail.

It measures about 140 microns long and 50 microns wide. (0.0055 x 0.002 inch)

Charlie--

That's just a spectacular shot. Its difficult to find a nice specimen of a Euglypha too. The local creek is running pretty high right now with all the rain, but this has inspired me to get my boots on and go grab some goop.

Beautiful work Charles. As usual though, I went off to Google this guy and learned it's shell is a test. That led me off on another search to find out just what biosilica is and how it's made. Not sure I understood everything, but I know more than I did before. :)

Hello Charles!

Very interesting for me. I have never seen the two "arms" of Euglypha brachiata. I am also thinking (like David) to excavate my boots!

I'm intrigued in how can you obtain so little focusing steps.

Thanks all.

A little embarrassing detail about the search...
After much time filtering and sifting through the collected matter I had finally found a nice specimen.
After cautiously transferring a drop of water containing the amoeba to a slide, I clumsily and accidentally flipped the slide over on my workbench. ](*,) :smt010

Fortunately, my workroom is situated in a part of the house where my wife could not hear my vivid vocalizations.

So I went back to the stereo and the sifted material and was fortunate enough to find this one. \:D/

Arturo,

I used Zerene Stacker. As I mentioned, due to the transparency of the test and the desire to retain the interesting surface detail, it was necessary to "break" the stack into 4 sections and then combine those four images. I did the final combination in Photoshop.
