Re: An aster floret cross sections
another wow!... excellent rendering.
Re: An aster floret cross sections
That calcofluor doesn't come cheap!
But I guess you don't have to use much of it?

Re: An aster floret cross sections
Really stunning! Can you tell us something about the optics and lighting?
Also, pardon my ignorance, but what is PBS and RT?

Re: An aster floret cross sections
Thank you!
the images were taken with a fluorescent microscope using lasers as a source of excitatory light - you may find more info on confocal microscopy here: https://en.wikipedia.org/wiki/Confocal_microscopy .
PBS = phosphate-buffered saline; a solution of salt (sodium chloride) at physiological concentration (~0.9%), buffered (Ph-stabilized) with sodium and potassium phosphates.
RT = room temperature (20 degrees Celsius, 68 F).

Thank you. Ii didn't realize they were done with a confocal microscope.
Re: An aster floret cross sections

I rarely look in the microscope forum, but now I'm glad I did. Thanks for sharing your technique and those stunning pictures.
