Hello David, Some nice work there! It appears you are quite happy with the PE wax and it sure seems to be doing the job. Did you need to use 100% EtOH? I have to get moving on thin sections - have everything but time. I picked up another microtome in addition to the AO 820 - it's a no-name (at least...

Hello Bernhard,

Excellent photos and interesting subject matter.

I continue to be impressed with the quality of images you get with the CP990 and assume you are using the background subtraction program on these.

All the best,
Mike

Hello Ralf, Thanks for the input! I'm actually pleased with DF aspect, (done with the 0.80 DF condenser on the Orthoplan) just not happy with the focus/positioning of the Closterium. As you know, when working with live 'critters' there is a fine line between getting good positioning/flattening and n...

Closterium - DF

Hello all, Still working on my photomicrography setup and making progress, albeit slowly! This is a DF shot I made while checking the flash capability - had to crank the Vivitar 283 all the way up! http://www.photomacrography.net/forum/userpix/54_Img1965small_1.jpg Hard to get a 'clean' image due to...

Hello Bernhard,

I thought as much - I've used a similar using ImageJ that does a wonderful job of flat field subtraction. I was actually hoping to find a CP that did not have artifacts, as all the version I've tried (995, 4500, 5000) are 'afflicted' to a greater or lesser degree.

All the best,
Mike

Hello Bernhard,

Very nice shots!

I see you are using a CP990 - do you have any problems with artifacts from the lens?

All the best,
Mike

Mosquito Proboscis

Hello all,

While I'm doing insect mouth parts, here's the working bits of a female mosquito, including the antennae.

Kind of a 'quicky' with 8 images stacked with Combine ZM.

All the best,

Hello Ken & David, Thanks for the comments. Yep, pretty ugly anyway you look at it, but presents a lot of detail on different planes to test the various stacking programs. This is actually a cheap Chinese mount, appears to be a squash using either a Meltmount of something similar. All the best, Mike

Housefly Proboscis

Hello all, Finally getting back into microscopy and mostly settled on using my Nikon D200 mounted on the Orthoplan using a Nikon UFX II Camera adapter with custom mount. The relay lens I'm using is a Nikon HK 2.5X, objective Leitz Pl 10X. All exposures are controlled by flash duration as set on a de...

Hello Graham, Excellent shot with lots of detail! I like the dust - shows a bit of its having spent untold hours swinging back and forth, dutifully counting off the seconds. It appears the weights on the balance wheel are of two different metals, it that correct? Amazing how much skill and technolog...

Hello Walter, Have to admit I can't figure it out; your shots look very much like those obtained with fluorescence setup, perhaps not as bright on the various fluorophores, but very close! From the NikonMicroscopyU website: "Applying a popular triple fluorophore technique, the BPAE cell culture pres...
