Even though I have only worked with magnificcations up to around 2-4x I see some problems in your setup. 1) Your object should be fixed as solid as your camera. Your nice, long wood sticks have lots of ressonance. Give it a lilttle knock at the end and it will continue to vibrate for several seconds...

From first look I felt it looked like a plant gall. Looking at the last picture I am certainly sure. A gall is a structure created by a host plant as response to an egg from a gall insect placed in a shoot, a budd, a leaf or a stem. Growth hormones from the egg (or perhaps from the mother?) interfer...

It is not difficult to find a manual for preparing some petri discs with sterile agar and then incubate them in differnt ways: Fingerprints, tissue used for different cleaning purposes, a little soil, etc. And of course remember a blind sample. Then keep them under cover at around 30 degrees Celsius...
