Lighting makes the biggest difference. Do you have some photos of your setup in use? I'm having trouble visualizing it. Also, do you use a stackshot or other automated focus rail? I'm currently using a sunwayfoto mfr-150s and while it's better than handheld for accuracy, I'm finding it very tedious...

Jesse, It sounds as if you are changing the f/stop on your zoom lens. Is this really the case? If so, we need to clear up some confusion. When you mount a microscope objective on a lens such as your zoom, you are using the zoom as a "converging lens" or "tube lens." Stopping down the converging len...

That ladybug image was taken with good diffused flash lighting and at a different angle. Probably generously and carefully sharpened as well. Not really comparable to your LED lighting. I was wondering about that. You think better lighting would make a huge difference? Where can I find a tutorial o...

Here are some example photos I just took of a ladybug. I'm using an LED light panel for illumination. 1/160 F11 ISO 1250: https://i.imgur.com/ZOjeDY3l.jpg 1/160 F8 ISO 500: https://i.imgur.com/xgpMlT3l.jpg These are cropped to remove the majority of the image circle since this objective doesn't play...

In search of greater depth of field
Hello, I bought a Nikon E400 with APO objectives last year and I've been enjoying it. I spent a lot of time and money setting up a Nikon f-mount adapter for video and photography, then I spent some more time and money playing with the 4x APO objective on-camera, on the end of my 70-200mm zoom lens. ...

That looks frustrating. This may be an "ignorant" question, as I have little experience; have you eliminated the projective eyepiece? (rotate 90°?) If by "projective eyepiece" you mean the relay lens in the trinoc tube, then yes, I've rotated it with no result. Also the dust speck shows up in both ...

Here's the tear down video if anyone is curious. I'm linking to 7:36 where the field lens has already been removed. Dusty in there. https://youtu.be/g-W9CEiEpPA?t=7m36s

After editing the footage... it could still be the field lens. It looks to be in pretty bad shape, even after cleaning. It looked clean to the naked eye, but the camera had a much better view. I think the next thing I try will be to rotate the field lens. I'll need to strip the microscope down to do...

If it is in the field lens it must focus and defocus moving the condenser knob. The method to find dirt is to rotate and focus each movable element Well, this is frustrating. The dust speck does indeed focus and defocus using the condenser knob. I had a moment, finally, to tear the microscope down ...

I just take a WB with no slide or with a blank part of the slide, or -more often- I shot raw an just use the eyedropper in a blank or neutral zone during conversion. Be aware that changing the halogen lamp intensity it changes colour temperature I shoot raw for stills, but I don’t have quite as muc...

Re: DSLR white balance
What is the correct way to whitebalance and meter a brightfield scene? Depending on the body you shoot something neutral a grey card or white card I have a grey card, but I'm not sure how you'd use one under a microscope. Would help if you mentioned that it was a microscope from the start, maybe in...

Finding dust speck that does not move
Hello, When using a camera to record video with my Nikon E600 microscope via the trinocular port, I see a dust spot on my camera that does not move when I move the stage. At first I thought it must be on the camera sensor, so I cleaned the sensor using Eclipse cleaning fluid and sensor swabs. I coul...
