Re: SEM scanning electron microscope protocol for diatoms and radiolaria

Hello Mark,

Thank you very much! I got you private message and have emailed you.

I am specifically interested in what reagent is used for gluing diatoms to cover slips and what is used as coat (I heard gold may not be a good choice).

I really appreciate your help!

With best wishes,
Frank

SEM scanning electron microscope protocol for diatoms and radiolaria

Dear forum friends, I may get temporary access to SEM (scanning electron microscope) to play with and am thinking to image some cleaned diatoms (kindly given by a forum member) and radiolaria. Samples are cleaned (in skeletons/ frustule form), loose, suspended in alcohol and not mounted. Can anyone ...

Re: What you wanted see in Scanning Electron Microscope?

Hello maikl36911,

I got temporary access to SEM as well, and would like to image some cleaned diatom frustules and cleaned radiolaria.

Would you please share a suitable protocol for cleaned diatoms? Thank you!

I can be reached at zzffnn at h0tm8il d0t c0m

Best,
Frank

Re: Microscopy method comparison: Triceratium grande Diatom

Macro_Cosmos, Haven't you tried your Leitz Heine in darkfield mode? With that you can get darkfield in objective NA of 1.1, depending on diatom (thinner ones with cleaner background respond better). Super thin diatoms, such as Amphipleura pellucida, surely "dot" better under oblique (COL) + pol than...

Re: 20mm Angel Eyes Ringlights

I have some thoughts / questions / comments on these LEDs: Is heat sink necessary, if the LED is turned on for less than 8 seconds each time? Or maybe one can simply thermo-glue the LED to a flat metal washer (granted, such heat sink is not optimal)? I like its flat and small profile, which is what ...

Re: 20mm Angel Eyes Ringlights

Wow, those 20mm and 40mm ones can be very useful for supermacro and micro. Especially when coupled with thin flexible wiring and battery power.

I don't have skills to rig them, but would be very interested to buy a few, if a member here doesn't mind sharing.

Re: Is this Chromatic Abberation? Lens flare?

Those are CA. Darkfield makes CA more obvious, compared to other illumination techniques. I would like to know if your immersion darkfield condenser's height is set optimally (move it up and down to see if that makes a difference) and if your condenser is properly centered. If not, you will get quit...

Re: Very bad image quality when using polarizers on a microscope, why?

Analyzer not being flat or not being big enough will definitely degrade image quality.

So with 27mm filters in/near eyepiece, you don't have significant image quality degradation anymore?

Re: Wanted: Heine Phase Contrast Condenser

Sorry, I missed your WTB by a few weeks, otherwise I would have sold mine to you. Mine went onto eBay and got sold the next day. Any reason why you really want it in your life :) I can ask around and see if I can find you another one. I don't think most people really need it though, if they already ...

Thank you, Jason. You saved the the event and made it more enjoyable. Other presenters there don't seem to remember that this is about "science meets art". There is artistic beauty in biology to be revealed. Even to me, as an immunologist and microbiologist PhD, this year's presentations have too mu...

Re: Mitutoyo 5x new open box $300 on ebay

Thanks for the tip. I have a 5x but it gives a noticeable amount of axial CA so I've been wanting to replace it. Hopefully I won't end up with two! Might be best in this case to buy two of these and test them. For that price you can always sell the extra lens. I've done that before with used eBay o...
