Yep, control for temperature, oxygen in the water and all of this while not moving the subject too much. Also high depth of field suggests some stacking.
The first few divisions when you see ripples in cells are something incredible.

Yep. If it has no means of changing focus position it will not work. But should work for microscope lenses or any other lenses on a mechanized platform.

Be careful with that thing. I was once bitten by one and was (almost forcefully) hospitalized for a short period of time to receive anti-rabies vaccine. Seems like no one cares if a stray dog bites you, but if it's a bat - then you're in trouble.

Thanks, zzffnn!

I used a concave slide and hydra was sitting close to its edge. Squeezed, but not too hard. 0.95x objective is a pain to work with. Working distance is next to nothing. But when it does work It gives very nice results.
