Technical notes
I was inspired by a cover story in the new issue of Microscopy Today about the opportunities stoma provide for student microscopy, So I gave it another shot today.
I took a leaf from a Christmas Cactus and carefully made a shallow incision in a rectangle, being careful to only cut the skin of the leaf. Then I peeled the patch up with a forceps.
The first attempt, I mounted the patch under a cover slip with a water mount. The main problem was particles floating through during the stacking process that I new would show up in the final shot.
I unmounted the patch, washed it and then made a glycerin mount. The viscosity stop particle flow significantly. 60X objective. Zerene Stacker.
