ICs with a 100X oil objective

Post by Olympusman »

This is fun stuff. The crazy colors are due to a WHITE BALANCE button in the TOUPVIEW software that comes up with these crazy colors. At times, I use a gray card to balance the scope, but I find these variations quite dynamic.

Really does bring out the detail. Very nice. Is that with a coverslip between the chip and objective, or oiled direct to the chip?

100X Oil

Post by Olympusman »

No cover slip - oil directly on the chip. When I frame the shot the image is very murky, but when I add the oil and get the meniscus the contrast jumps dramatically.
I save my chips on slides, and after my sessions I clean them in xylol.

I wonder if you might get even more contrast/sharpness with a coverslip too. I think I remember your 100x oil being a 160/0.17 - so the coverslip is designed in. Even if the oil has a very close RI to the glass, it should make a difference. Perhaps a thin film of oil between the coverslip and the chip too - just to keep it as homogenous as possible.

Cover slip

Beatsy,
I'll give it a try, but the major thing that scares microscopists aout oil immersion is the excessive cleanup. I have worked out a very efficient cleanup system, so we'll see how it goes.
