Diatom under interference contrast illumination

Post by Brian Matsumoto »

I have spent the last two years working on my Zeiss microscope with Jamin-Lebedeff optics. It has been a struggle, but I have been pleased with the effort in learning a new form of illumination. Getting the hardware to work for generating images took some effort. One of the features of this form of illumination is that the colors can be varied and I am posting two images made at different settings. The pictures while of the same specimen (Arachnoidiscus ehrenbergii) are not comparable as the one with the red background was photographed on a Zeiss Photomicroscope while the one with the blue background was taken on a Zeiss Universal. I suspect the Universal may have had a misaligned Optovar component. But now I think I can now spend my time taking pictures with the setup. The images are an extended depth of field pictures.
