There's no hard-and-fast number. I've seen good results from 20-ish frames. More important is the physical aspect ratio of the subject, depth:width. Use the formulas at https://zerenesystems.com/cms/stacker/d ... wing_angle to calculate what shift % you need, based on your subject dimensions and viewing angles.

Thanks for the additional information on viewing angle and step estimation Rik. I need to have another go at imaging the nucleus.
