Although the non-diffused lighting is quite harsh with white highlights, I'm not sure I prefer the diffuse lighting. Which works best for you?

Nicely done. I generally prefer the effect of diffuse lighting, but in this example the specular highlights and shadows in the first image make it look more detailed. I wonder if the red channel is a bit overexposed in the second image?
As with most things though, I suspect the best answer is somewhere between the two (partially diffused light with less falling on one side if the specimen).
