I have been playing with my fluorescence set up recently. To date I have built a simple 1W blue LED lightsource and I have made my 445nm laser set up safer. I have a new laser module on a heatsink with a low power lens so the beam diverges nicely when I plug it into the lamp holder on my Olympus microscope. I use dark ground illumination and a 450nm or 500nm blocking filter. The advantage of using a laser is that it is monochromatic so even the 450nm blocking filter gets rid of almost all of the stray blue light. Anyway here are a couple of the preliminary images.

1. is an old prep of daffodil pollen (unstained).

2 & 3 are bacteria stained with acridine orange.

The images were photographed using an EOS 6D at 1600 ISO and exposures of 4 to 8 seconds.

These are very interesting, thanks for sharing.

I was thinking of having a go at something similar myself. Could you give us a bit more information about your blue light and cost/where to buy in UK?
