diatom comparison

Post by mtuell »

Here is that same Victorian mount diatom I showed in stereo. 80 micron diameter.

40x Ph3 160/0-2 objective. Nikon D3300 on Diaphot TMD. Phase Contrast LWD 0.52 condenser. Zerene stacks of ~15 images.

This time, brightfield with a 60x Plan Apo /1.4 (stopped down) with a 1.25 Phase Contrast condenser (oiled).

90 image DMap Zerene stack with Photoshop post-processing. First, a 33% sized version, then a 100% crop.

And, with a slightly deeper stack, some fun Photoshop magic!

This is the Victorian mount slide, so it gets difficult to do all the diatoms on there at high NA, since they may not be at the same depth.
