No staining.
I used a somewhat different approach for this picture. A live leaf with a drop of microscope oil (no coverslip), a Nikon plan apo 60x Na 1.4 oil 160mm lens and Epi light (!).

This was with the EPI light for 210 mm tube so I could not use the trinocular head because the tube length would have been to long. I have also been experimenting with "normal light" trough a fluorescens EPI light and a polarizing cube, It is ok for viewing.
It would bee nice to have a high NA infinity water dipping lens.
