How do I calculate the correct tube lenghth?

I am trying to put together a good setup with a microscope lens on my
Nikon 800e. I want to use extention tubes, but how do I calculate the correct
tube distaance? For infinity lenses, 160 lenses?
What is the best possible lenses to use like this for extreme macro?

For more specific information, you'd need to tell us exactly which objective(s) you want to work with.

There are different things to take into account for:
- optics requiring compensating eyepieces vs. CF objectives
- pre-DIN and DIN objectives
- finite and infinite tube length
- full frame (as you have) vs. crop sensor cameras
- cameras with EFSC and those without (this is going to be an issue for you)

Yes.
Otherwise the quality will suffer.
With lower magnification, and lower NA, it's less important.
