Just guessing on this diatom. Anyone happen to know? Perhaps a link. I wasn't able to find it in the database I was looking at.

Another diatoms today. <g> I'm not sure how to capture the 'sphere' stacking wise. Either the objective won't resolve it properly, or I need to change some settings in Zerene. I haven't a clue how to stack any smaller increments if that's the problem. In any case at least you get the illusion of it being a sphere even though it gets less sharp as you reach the circumference.

The dry objective is likely the cause of the "out of focus circumference". As you focus "deeper" into the mountant, the correction for coverslip thickness drifts out of "optimum". Same happens with non correcting objectives at lower NA, just not quite as quickly.

If depth of mountant is the problem, then you might be able to work around it by pausing the shoot midway and re-adjusting the correction collar.

There's no guarantee this will work, however. Features along the periphery of a sphere may be seen by only about half of the objective's aperture, while the other half of the aperture sees OOF foreground. This effect can cut contrast pretty badly. It's another version of the issue diagrammed at http://www.photomacrography.net/forum/v ... 042#135042 .
Either the objective won't resolve it properly, or I need to change some settings in Zerene.
Check your source images. If source is sharp, then it make sense to think about changing settings in Zerene. If source is not sharp, Zerene cannot fix the problem.

Thanks to you! I'm having a blast. I can spend 'weeks' on these puppies. I've photographed several dozen already, just haven't posted them. Didn't want to bore anyone. <g> I can't wait to figure this all out and buy a couple high-end objectives.
