Why photo-stack ?
Why is it that we go to all the trouble of photo stacking when we see TV documentaries whereby the tiniest of insects can be photographed (even in motion) perfectly sharp using the special lenses developed by that chap (his name escapes me) that David Attenborough endorses or are those images someho...

Re: Nikon "U" objective aperture
I'm curious as to why they close down so far. Woudn't it simply be to get best depth of field based on the premise that the objective may not have been designed with living subjects in mind. So either strong illumination or extended exposure times could be used. And it was probably designed long be...

Second Hand Nikon Instrument-very solid.
I have recently purhased on ebay a Nikon microscope-I think a S series polarizing instrument from the 1970's. I intend to set it up on a fixed stand, probably a modified enlarger column that incorporates a camera and bellows attachment. I am currently building a small house so am a bit preoccupied a...

Yes such a stage would be nice. I did see one on ebay (rather expensive) but it lacked an x-y adjustment for the slide. Without such fine control I would never be able to find the subject. I actually purchased off Ebay a Nikon microscope just the other day with the rotating stage with no x-y adjust...

Re: Improving Depth of Field through the microscope.
I have downloaded a trial version of Zerene but it seems that one cannot save output images on the trial version. Is that the case ? The 30-day trial period is fully functional, no restrictions. To save an output image, be sure the image you want is selected (highlighted), then in the menu system u...

Hi Charles. Thanks for the welcome. You said There's still place for an "inserted" aperture in a compound microscope. But I think it's biggest value would be in situations where you are trying to establish darkfield illumination with an objective whose numerical aperture is a little too large for th...

Georgetsmurf intro
Hi. I thought I better say a bit about myself as I have only just joined the site. I have been photographing one way or another since about 1963. Those with a flair for mathmatics could work out how old or young I am from that hint. I have done quite a bit of general macro stuff but not much through...

Re: Improving Depth of Field through the microscope.
You say Stopping down is not a good path to increased DOF in this regime. Your time will be better spent learning about focus stacking. --Rik I have downloaded a trial version of Zerene but it seems that one cannot save output images on the trial version. Is that the case ? I have put a trial stack ...

Thanks for that-please point me in the right direction and as Dick Tracy (i think) would say-"I will get on it right away".rjlittlefield wrote:Stopping down is not a good path to increased DOF in this regime. Your time will be better spent learning about focus stacking.

These images look good -- sharp and no obvious CA (chromatic aberration, color fringes). Next step: focus stacking. I am curious about your equipment. Are you using flash illumination or continuous? How do you have your camera coupled to your scope? --Rik These images were from a camera (Nikon D300...

Improving Depth of Field through the microscope.
As with all photography-to getter better results there is always a trade of to be addressed. I am very new to Photomicrography other than some stuff for uni many years ago. I have however done and published quite a lot of macro work. I have found that to imrove depth of field through the microscope-...

Dark-field modification
Sorry for my ignorance but new to this discipline. How does one set up a conventional microscope for Dark-field photomicrography?
